In order to examine the structure of ancient parchment specimens, small fragments were softened in a solution of trypsin, were embedded in paraffin and cut laterally and transversely with a rocking microtome, mounted, and viewed under the microscope. The author describes the various skin layers present and the difficulty in species identification of the papillary, or grain lay has been scraped away as was often common in 13th century England.
