Abstract
Isoelectric focusing is used to characterize proteins from various types of animal skin and collagen products that have been used to manufacture books and manuscripts. Identification of parchments is difficult if the grain layer is scraped away during manufacture of the material. Isoelectric focusing can be used to identify the animal source to the genus level. With further research the method can be used to age parchments based on a correlation of hydrolytic protein products with shrinkage temperature. The isoelectric focusing method operates by creating a pH gradient in an electric current. Dissolved collagen from the samples is placed on the gradient and the current is applied. The amphoteric components of the proteins (i.e. the component amino acids) migrate towards their respective isoelectric points and stop. The technique can resolve substances to 0.001 pH units. The equipment used was an LKB 2117 Multiphor Electrophoresis chamber using an LKB 2130 power supply and an LKB Multitemp II Thermostatic circulator set at 6°C. Polyacrylamide gel (PAG) plates were the substrate. The parchment samples were obtained from the De Vrijenban Company, Rotterdam, Holland. The outcome of this study was to supply visual data of the focused protein bands for comparison with unknown samples and to serve as a preliminary study for the aging of parchments.
