Abstract
A standard protocol is presented using an enzyme-linked immunosorbent assay (ELISA) for the simultaneous identification of binding media from sources of mammal (collagen from skin or bone glue; casein from milk); avian (ovalbumin from egg white; phosvitin from yolk), and plant (polysaccharides from gums). The ELISA results showed that different proteins and polysaccharides are identified even in mixtures or together with other binders (oils, wax, and resins) as well as in the presence of pigments. Further experiments using reference binding media verified that the detection limits for the assay are extremely low, on the order of nanograms. Protein and gum-based binders from many animal and plant species were tested by ELISA, and the results indicate that carefully chosen antibodies detect these binding media from most natural sources.
